Stacofylline (S 9977 free base; S 9977-2 free base), a Xanthine (HY-W017389) derivative, is a potent AChE inhibitor with an IC50 of 5-50 nM. Stacofylline shows anti-amnesic and promnesic activities[1][2]. In Vitro:Stacofylline potentiates cholinergic transmission by significantly increasing the time constant of decay (Tc) of inhibitory postsynaptic currents (IPSCs)[1]. In Vivo:Stacofylline (0.0312-0.5 mg/kg; p.o.) clearly attenuates the memory deficits induced by the three amnesic treatments after acute treatment in Scopolamine, Diazepam, and electroconvulsive shock (ECS) mouse models,[2].
