VJ115

VJ115 is a potent ENOX1 inhibitor with an EC₅₀ of 50 μM. By inhibiting ENOX1, VJ115 elevates NADH, which in turn downregulates the phosphorylation of p70S6K1/S6, alters the expression of c-Jun and cytoskeletal proteins, induces endothelial cells to lose polarization and migration ability, and increases caspase-3-dependent apoptosis under radiation. VJ115 can be used in studies related to solid tumors^[1][2]. In Vitro:VJ115 (50 μM; 2-6 h) increases the intracellular NADH fluorescence level, inhibits the formation of capillary-like tubular structures, downregulates the mRNA and protein expressions of lamin A/C and stathmin, upregulates the mRNA and protein expressions of Hup1, and reduces the protein levels of phospho-cJun and phospho-p70S6K in HUVEC cells^[1]. In Vivo:VJ115 inhibits developmental vasculogenesis in zebrafish embryos, phenocopying the effects of ENOX1 suppression^[1].
VJ115 acts as a radiation sensitizer, reducing tumor microvessel number and density and delaying syngeneic and xenograft tumor growth when combined with fractionated x-irradiation, while enhancing radiation-induced endothelial cell apoptosis^[1].
