Lysipressin acetate (Lysine vasopressin acetate; [Lys8]-Vasopressin acetate) is a neurohypophyseal nonapeptide with dual cardiovascular regulatory activities of peripheral pressor action and central hypotensive action, and it also regulates pain perception in the body. Lysipressin acetate is an Oxytocin Receptor agonist, with a Ki value of 10.2 nM for porcine OT receptor. Lysipressin acetate stimulates contraction of rabbit bladder smooth muscle via V1-vasopressin receptor. The central hypotensive effect of Lysipressin acetate depends on the sympathetic tone pathway. Lysipressin acetate dose-dependently prolongs tail-flick latency in rats, producing a stable central antinociceptive effect. Lysipressin acetate can be used in studies related to cardiovascular regulation, hypertension and pain[1][2][3][4]. In Vitro:Lysipressin (LVP) acetate enhances the contractile capacity of uterine muscle strips from luteal-phase and prepartum pigs in a dose-dependent manner by activating oxytocin receptors, with EC50 values of 0.392 μM and 0.223 μM, respectively; it also increases intracellular Ca2+ concentrations in prepartum porcine uterine myocytes, with an EC50 of 28.2 nM[1].
Lysipressin acetate dose-dependently displaces [3H]oxytocin binding sites on myometrial membranes of luteal-phase pigs, with a Ki value of 10.2 nM, indicating its high affinity for oxytocin receptors[1].
Lysipressin (LVP) acetate stimulates the contraction of bladder smooth muscle from male New Zealand white rabbits in vitro, with a mean pD2 value of 8.4[2]. In Vivo:Lysipressin (Lysine vasopressin) (0.01-100 μ/kg; intravenous injection; single 5-second injection; intracisternal injection; single 5-second injection) acetate induces dose-dependent peripheral pressor responses and bradycardia, as well as dose-dependent central hypotensive responses with minimal heart rate changes in anesthetized normotensive dogs[3].
Lysipressin (1.0-10.0 μ/kg; intravenous injection; single 5-second injection; intracisternal administration; single 5-second injection) acetate loses central hypotensive activity in anesthetized dogs after Guanethidine (HY-B1251) pretreatment, whereas its peripheral vasopressor effect stays unaltered[3].
Lysipressin (150-500 ng; intracerebroventricular injection; single administration) acetate produces significant, dose-dependent analgesic effects in the radiant heat tail-flick test[4].
