Pregnanediol (NSC 1612) is a Progesterone (HY-N0437) metabolite. Pregnanediol does not affect the transcriptional activity of UGT1A1 enhancer-promoter complex of WT and variant type. pregnanediol inhibits glucuronidation activity of G71R-UGT1A1. Pregnanediol is a cause of breast milk jaundice in carriers of G71R[1][2].
In Vitro:Pregnanediol (0-100 μM) reduces bilirubin glucuronidation of G71R-UGT1A1 in COS-7 cells, decreasing the bilirubin glucuronidation activity of G71R-UGT1A1 to 51% of that in the wild type at 100 μM[1].
Pregnanediol (0.01-100 μM) does not affect the transcriptional activity of UGT1A1 enhancer-promoter complex of WT and variant type in HepG2 cells[1].
