11alpha-Hydroxyprogesterone

11alpha-Hydroxyprogesterone is a naturally occurring steroid hormone and an 11β-hydroxysteroid dehydrogenase (11β-HSD) inhibitor. 11alpha-Hydroxyprogesterone decreases FAK protein expression and regulates epithelial-mesenchymal transition, cell migration, and cell invasion. 11alpha-Hydroxyprogesterone exhibits antiandrogenic and blood pressure-regulating activities. 11alpha-Hydroxyprogesterone is used in the research of breast cancer and malaria^[1][2]. In Vitro:11alpha-Hydroxyprogesterone (11α-OHP) (1-10 µM; 24 h) inhibits the migration and invasion of MDA-MB-231 cells^[1].
In MDA-MB-231 cells, 11α-OHP (10 µM; 24 h) treatment directly increases E-cadherin levels and reduces vimentin expression^[1].
11α-OHP (1-10 µM; 24 h) does not exhibit cytotoxic or proliferative effects on MDA-MB-231 cells^[1].
11α-OHP treatment does not significantly alter E-cadherin or N-cadherin expression in MCF-7 cells^[1].
11alpha-Hydroxyprogesterone is a high-affinity hit compound that binds to the active sites of both Plasmodium falciparum FabI and FabZ, with binding energies of −10.7 kcal/mol and −10.0 kcal/mol, respectively^[2]. In Vivo:11alpha-Hydroxyprogesterone (11α-OHP) (60 mg/mouse; s.c.; over a 5-week period) does not affect primary tumor growth but significantly suppresses lung metastasis in the MMTV-PyMT breast cancer model by modulating EMT markers^[1].
