2′-Deoxy-2′-fluoroguanosine

2′-Deoxy-2′-fluoroguanosine is a nucleoside analog and potent influenza virus inhibitor with an EC₉₀ value of <0.35 μM against influenza A and B viruses. 2′-Deoxy-2′-fluoroguanosine inhibits influenza virus replication in the upper respiratory tract, thereby ameliorating fever and nasal inflammation^[1][2][3]. IC50 & Target:EC90: <0.35 μM (influenza virus A and B)^[1] In Vitro:2′-Deoxy-2′-fluoroguanosine (10 μM) blocks primary and secondary transcription of influenza virus X31 strain RNA in chicken embryo cells^[1]. In Vivo:2′-Deoxy-2′-fluoroguanosine (5-40 mg/kg; i.p.; 1-3 times, administered at 1 h, 1 and 24 h, 1, 8 and 24 h after infection, respectively) can reduce the viral load in the upper respiratory tract, alleviate fever and inflammation in the ferret model of influenza A virus clone 7a infection with a single dose, and 3 doses can reduce the viral load in the lung by > 100-fold^[2].
