Monochlorobimane (Chlorobimane) is a fluorescent dye (λex=380 nm, λem=470 nm) to measure glutathione (GSH) in cellular assays[1].
In Vitro:Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Solution preparation[3]
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 10 mM.
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute to 1-10 µM with PBS or cell culture medium (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Cell staining
2.1 Cells are plated in clear-bottomed black 96-well microplates.
2.2 Add Monochlorobimane (1-10 μM) and incubate at 37°C for 30 minutes (optimized according to the experiment).
2.3 The fluorescence intensity is measured with fluorescence microscopy or Fluorescence microplate reader (Ex/Em = 380/480 nm).
