Aβ aggregation modulator-1 is a stimulator of amyloid-β () fibrillogenesis. Aβ aggregation modulator-1 binds hydrophobic residues in peptides and stabilizes β-sheet-rich protofibrils and fibrils. Aβ aggregation modulator-1 accelerates polymerization and reduces concentrations of small, toxic oligomers in heterogeneous aggregation reactions. Aβ aggregation modulator-1 suppresses long-term potentiation (LTP) inhibition by oligomers in hippocampal brain slices. Aβ aggregation modulator-1 can be used for the study of Alzheimer's disease (AD)[1]. In Vitro:Aβ42-IN-6 (Compound O4) (48 h) reduces Aβ42-induced toxicity in differentiated SH-SY5Y cells and primary cortical neurons[1].
Aβ42-IN-6 (5-25 μM; 48 h) promotes aggregation of Aβ42 in SH-SY5Y cell culture media, decreasing the abundance of soluble Aβ42 monomers and small oligomers while increasing larger aggregate structures[1].
Aβ42-IN-6 (5 μM, 48 h) stimulates the formation of SDS-resistant Aβ aggregates in 7PA2 cells (CHO cells overexpressing mutant human APP)[1].
Aβ42-IN-6 suppresses Aβ42- induced inhibition of hippocampal long-term potentiation (LTP) in the CA1 area of rat brain slices, rescuing the dysfunction of synaptic plasticity caused by toxic Aβ42 oligomers[1].
