Swertiajaponin is a tyrosinase inhibitor, forms multiple hydrogen bonds and hydrophobic interactions with the binding pocket of tyrosinase, with an IC50 of 43.47 μM. Swertiajaponin also inhibits oxidative stress-mediated MAPK/MITF signaling, leading to decrease in tyrosinase protein level. Swertiajaponin suppresses melanin accumulation and exhibits strong anti-oxidative activity[1].

In Vitro:Swertiajaponin (2-20 μM, 48 h) shows no cytotoxicity in vitro[1].
Swertiajaponin inhibits melanogenesis in human skin model (10-20 μM, 5 days) and UVB or αMSH-induced melanogenesis in B16F10 cells (2-10 μM, 49 h) in dose-dependent manner[1].
Swertiajaponin (2-10 μM, pretreated for 1 h and then UVB exposure for 30 min) decreases ROS and ONOO- in concentration-dependent manner[1].
Swertiajaponin (2-10 μM, 7 or 49 h) inhibits UVB-induced MAPK activation[1].
