INK4C-IN-2 is a p18INK4C inhibitor. NK4C-IN-2 promotes hematopoietic stem cells (HSCs) division by inhibiting INK4C and activating CDK4/6, with an ED50 of 5.21 nM. INK4C-IN-2 shows no cytotoxic to normal 32D cells, HSCs, leukemia cell lines and myeloma cell lines[1].
In Vitro:INK4C-IN-2 (Compound 40) (0.01-1 μM, 7 days) promotes human HSC expansion ex vivo[1].
INK4C-IN-2 (14 days) increases number of Phenotypic Long Term (LT)-HSCs[1].
INK4C-IN-2 (5 days) stimulates HSCs division by inhibiting INK4C[1].
In Vivo:INK4C-IN-2 (Compound 40) (Culture CD34+ LKS cells with 20 μM for 3 days, i.v., 16 weeks) increases the functional LT-HSC populations in competitive bone marrow transplantation mice engraftment experiments[1].
