Protohypericin is a compound that can be extracted from Hypericum perforatum. Protohypericin has low photocytotoxicity but can be efficiently photoconverted into Hypericin (HY-N0453) with high phototoxicity under visible light. Protohypericin can be used in cancer research[1][2][3][4].
In Vitro:Protohypericin (80-200 μM; 3-5 h) exhibits extremely low transepithelial transport rate and significant intracellular accumulation in Caco-2 cells[1].
Protohypericin (0.25-1 μM; 24 h incubation + 15 min irradiation) can induce photocytotoxicity in HeLa cells with a CC50 of 0.21 μM[2].
In Vivo:131I-labeled Protohypericin can target tumor necrotic regions. When 131I-Protohypericin is combined with the vascular disrupting agent Fosbretabulin (HY-14815), it exhibits antitumor activity in a non-small cell lung cancer mouse model[3].
