Desogestrel (Org-2969) is a third-generation progesterone analogue contained in many oral contraceptive preparations. Desogestrel is a click chemistry reagent, it contains an Alkyne group and can undergo copper-catalyzed azide-alkyne cycloaddition (CuAAc) with molecules containing Azide groups[1][2][3][4]. In Vitro:3-Ketodesogestrel (HY-B0652) is the active metabolite of Desogestrel. 3-Ketodesogestrel decreases PHOX2B expression by binding to the progesterone nuclear receptor PR-B[5]. In Vivo:Desogestrel (0.0083-0.083 mg/kg, po, every 4 days for 1 month) reduces serum levels of FT3, FT4, TPOAb and TgAb, upregulates TSHR and NIS mRNA expression, and stimulates the proliferation of thyroid follicular epithelial cells in Wistar rats models[6].
