Desacetylcinobufotalin is an active component isolated from Venenum Bufonis, which exhibits significant cytotoxicity against human hepatocellular carcinoma HepG2 cells (IC50 = 0.0279 µmol/mL). Desacetylcinobufotalin upregulates Bax protein expression, downregulates Bcl-2 protein expression, and induces apoptosis via the mitochondrial pathway. Desacetylcinobufotalin inhibits cancer cell survival, and shows lower cytotoxicity compared to its parent compound Cinobufagin (HY-N0421). Desacetylcinobufotalin can be used in hepatocellular carcinoma-related research[1][2].

In Vitro:Desacetylcinobufotalin (24 h) potently inhibits the survival of human hepatocellular carcinoma HepG2 cells with an IC50 of 0.0279 µmol/mL after 24 h, showing dose-dependent cytotoxicity and inducing cytoplasmic membrane damage[1].
Desacetylcinobufotalin (DEBF) (0.005-0.1 µmol/mL; 24 h) induces dose-dependent apoptosis in human hepatocellular carcinoma HepG2 cells after 24 h, with late apoptosis reaching 45.15% at the highest tested concentration of 0.1 µmol/mL[1].
Desacetylcinobufotalin, produced via biotransformation of Cinobufagin (HY-N0421) by human intestinal bacteria, exhibits significantly lower activity and reduced cytotoxicity relative to cinobufotalin[2].
