JNJ 10329670 is a potent and selective noncovalent cathepsin S inhibitor with a Ki value of 34 nM for human cathepsin S. JNJ 10329670 blocks invariant chain proteolysis in B cells and dendritic cells, as well as antigen-induced T cell proliferation[1].
In Vitro:JNJ 10329670 (0-100 μM) inhibits cathepsin S and thereby block the proteolysis of the invariant chain in human B cell lines[1].
JNJ 10329670 (0-10 μM) blocks proliferation of T cells in response to both antigens, but less so to the nonspecific T cell mitogen phytohemagglutinin[1].
