DAO-IN-2 is a d-amino acid oxidase (DAO) inhibitor (h-DAO IC50 = 245 nM). DAO-IN-2 is a moderate inhibitor of DAAO in vitro and in vivo. DAO-IN-2 can be used to study DAAO-related psychiatric disorders[1][2].
In Vitro:DAO-IN-2 (Compound 8) has no off-target activities for the inhibition of P450 enzymes (IC50s > 10 μM for CYP3A4, CYP2D6 and CYP3C9) or binding to ion channels[1].
DAO-IN-2 inhibits D-amino acid oxidase (DAAO) activity against expressing either the human or rat DAAO gene CHO cells, with IC50s of 144.6, 113.9 nM[2].
In Vivo:DAO-IN-2 (Compound 8) (10-200 mg/kg, i.p., once) inhibits DAAO activity in the kidney and cerebellum, increases plasma and cerebrospinal fluid D-serine levels, and reduces amphetamine-induced psychomotor spontaneous activity in Wistar rats model[1].
