ICA-105574 is a potent and efficacious hERG channel activator. The primary mechanism by which ICA-105574 potentiates hERG channel activity is by removing hERG channel inactivation. ICA-105574 steeply potentiates current amplitudes more than 10-fold with an EC50 value of 0.5 +/- 0.1 μM and a Hill slope (n(H)) of 3.3 +/- 0.2. ICA-105574 can prevent arrhythmias induced by cardiac delayed repolarization. ICA-105574 shortens action potential duration in ventricular myocytes concentration-dependently[1][2]. In Vitro:ICA-105574 (1 μM) reverses the inhibition of peak repolarizing currrents by Dofetilide (HY-B0232) and Moxifloxacin (HY-66011A) to the controlled levels in guinea-pig ventricular myocytes [2]. ICA-105574 (3-10 μM) induces concentration-dependent shortening of APD in guinea-pig ventricular myocytes[2]. ICA-105574 (3 μM) completely reverses the APD prolongation when combined with NS1643 (HY-16916) (10 μM)[2].
