BMS-341400 is an orally active selective phosphodiesterase 5 (PDE5) inhibitor with an IC50 value of 0.3 nM. BMS-341400 reduces the degradation of cyclic guanosine monophosphate (cGMP), thereby enhancing nitric oxide (NO)-mediated relaxation of the smooth muscle of the corpus cavernosum and promoting erection. BMS-341400 can be used to study erectile dysfunction.
In Vitro:BMS-341400 (Compound 6) effectively enhanced the relaxation of the smooth muscle in the isolated tissue of the rabbit penile corpus cavernosum, with its EC₅₀ value being 13 nM[1].
In Vivo:BMS-341400 (Compound 6) (10-1000 nmol/kg, i.v., single dose) effectively promotes the erectile response in the male rabbit model with EC₅₀ value of 100 nmol/kg[1].
BMS-341400 (10-250 mg/kg) demonstrates excellent safety in rats[1].
