Demethylsuberosin

Demethylsuberosin (7-Demethylsuberosin) is a coumarin compound found in Angelica gigas Nakai. Demethylsuberosin exerts antihypertensive effects by inhibiting the L-type Ca_V1.2 channel. Demethylsuberosin has antioxidant and anti-inflammatory activities. Demethylsuberosin also exhibits neuroprotective activities against glutamate-induced toxicity in primary cultured rat cortical cells^[1][2][3]. In Vitro:Demethylsuberosin (7-Demethylsuberosine shows good ABTS⁺ radical scavenging activity (IC₅₀ of 8.1 μg/mL)^[1].
Demethylsuberosin significantly suppresses NO production in Lipopolysaccharides (HY-D1056) (LPS)-stimulated RAW 264.7 cells, with an IC₅₀ of 7.6 μg/mL. Demethylsuberosin effectively suppresses the expression levels of iNOS, IL-1β, and COX-2, which are responsible for promoting the inflammatory process^[1].
Demethylsuberosin (100 nM-50 μM) shows a dose-dependent inhibition of Ca_V1.2 channel currents in HEK293T cells expressing the Ca_V1.2 channel, with an IC₅₀ of 3.23 μM^[2].
In Vivo:Demethylsuberosin (1-50 mg/kg; intragastric administration; once a day; 28 days) reduces blood pressure in L-NAME (HY-18729A)-induced hypertensive mouse models^[2].
