Introduction of 2101955-00-2 :

JAK2 JH2 Tracer (Tracer 5) is a fluorescent probe for JAK2 JH2 domain, with a Kd value of 0.2 μM[1][2]. In Vitro:Guidelines (The following is our recommended protocol. It is provided as a guide only and should be modified to suit your specific needs).
1.1 Preparation of storage solution
Use anhydrous DMSO to prepare 10 mM JAK2 JH2 Tracer stock solution.
Note: It is recommended to store JAK2 JH2 Tracer stock solution at -20℃ or -80℃ in the dark after aliquoting.
1.2 Preparation of working solution
Dilute the storage solution with preheated serum-free cell culture medium or PBS to prepare 1.5 pM JAK2 JH2 Tracer working solution.
Note: Please adjust the concentration of JAK2 JH2 Tracer working solution according to actual conditions and prepare it before use.
1 Preparation of dye working solutions[1]
1) Prepare a 1.5 pM JAK2 JH2 Tracer working solution, which is the minimum concentration that maintains a satisfactory signal-to-noise ratio.
2 Specific staining steps
1) Prepare a series of JAK2 JH2 protein solutions ranging from 0 to 4.8 μM in assay buffer.
2) Add 1.5 pM JAK2 JH2 Tracer working solution to microplate wells.
3) Add JAK2 JH2 protein solutions to the wells containing JAK2 JH2 Tracer stepwise to cover the desired protein concentration range.
4) Measure fluorescence polarization values over 90 min.
In Vivo:The possibility that mutations within the JAK2 JH2 domain may be associated with pathology, which is associated with a hyperactive kinase and a hematopoietic malignancy phenotype in flies[2].
Specific mutation in the JAK2 JH2 domain is associated with several myeloproliferative diseases[2].
