Kasugamycin (Ksg) hydrochloride hydrate is an antibiotic that binds to 30s and 70s ribosomes but not to the 50s subunit, and has anti-infective activity. Kasugamycin hydrochloride hydrate mimics mRNA nucleotides, disrupts tRNA binding and inhibits canonical translation initiation. Kasugamycin hydrochloride hydrate increases the sensitivity of mycobacteria to Rifampicin (HY-B0272) in vitro and in mouse infection models[1][2][3].
In Vitro:Kasugamycin hydrochloride (1 mM) inhibits P-site tRNA binding through destabilizing mRNA[1].
Kasugamycin hydrochloride (50 μg/mL) decreases the mistranslation rates of asparagine-to-aspartate and glutamine-to-glutamate[3].
Kasugamycin hydrochloride (0-100 μg/mL) reduces mycobacterial rifampicin phenotypic resistance in wild-type and high mistranslating gatA mutant mycobacterial strains[3].
In Vivo:Kasugamycin hydrochloride (400 mg/kg, i.p., 2 w) results in dose-dependent concentration-time profiles in plasma with rapid clearance of M. tuberculosis in lung burden of Mtb-infected mice[3].
