Cucurbitacin E is a CDK1 inhibitor that significantly inhibits the activity of the cyclin B1/CDC2 complex. Cucurbitacin E also induces PANoptosis in adrenocortical carcinoma cells in a ZBP1-dependent manner. Cucurbitacin E exhibits synergistic effects with Mitotane (HY-13690); when used in combination, they effectively eliminate tumors[1][2][3]. IC50 & Target:Cyclin B1/CDC2 complex[1] In Vitro:Cucurbitacin E (2.5-7.5 μM; 6-24 h) induces morphological changes in primary colon cancer cells and inhibits tumor growth by arresting the cell cycle at the G2/M phase through the induction of GADD45γ gene expression and the blockade of the Cyclin B1/CDC2 complex in primary CRC cells[1]. In Vivo:Cucurbitacin E (0.5 mg/kg) induces a significant reduction in body weight and a decrease in fat pad weight in HFD-MetS mice; specifically, total body fat mass is decreased by 55%, and the degree of central obesity is reduced to 50% of that of the control group[2].
