Segetalin B, an orally active cyclopentapeptide found in Vaccaria segetalis, possesses estrogen-like activity. Segetalin B promotes mineralization of ovariectomized rat-derived bone marrow mesenchymal stem cells (BMSCs) in vitro and increases the level of osteocalcin, BMP-2, ALP, and SIRT1 activity. Segetalin B is promising for research of post-menopausal osteoporosis (PMOP)[1][2][3][4].
In Vitro: Segetalin B (0.1-100 μM, 24 h) shows significant cytotoxicity at 100 μM[4].
Segetalin B (0.1-10 μM, 15 days) enhances the mineralization of ovariectomized rat-derived BMSCs and increases ALP activity and levels of Osteocalcin and BMP-2[4].
Segetalin B (10 μM, 15 days) upregulates the protein expressions of Runx2 and osterix in BMSCs[4].
In Vivo: Segetalin B (2.5 mg/kg, s.c., daily for two weeks) increases the uterine weight in ovariectomized rats, showing the estrogen-like effects[1].
Segetalin B (10-160 mg/kg, p.o., daily for 4 weeeks) inhibits bone loss in ovariectomized rats and upregulates expressions of Runx2, osterix, and SIRT1, and downregulates expressions of NICD and Hes1 in bone tissue of ovariectomized rats[4].
