para-Nitroblebbistatin is a derivative of Blebbistatin (HY-13813) and an inhibitor of myosin II. para-Nitroblebbistatin is photostable, non-cytotoxic, and non-phototoxic. para-Nitroblebbistatin can serve as an ideal substitute for Blebbistatin (HY-13813) to study the role of myosin II in physiology, development, and cell biology[1][2].
In Vitro:para-Nitroblebbistatin (0.31-10 μM; 24 h) can inhibit cytokinesis in COS-7 cells, resulting in a multinucleated phenotype in the cells, with an EC50 of 2.9 μM[1].
para-Nitroblebbistatin (10 μM; irradiated with 480 nM for 15 min; 18 h) is non-cytotoxic in HeLa cells, and there are no differences in cell mortality and morphology compared with the control group[2].
In Vivo:para-Nitroblebbistatin (5-20 μM; single dose; 24 h) can inhibit the cell migration of the posterior lateral line primordium in transgenic cldnb:gfp zebrafish embryos, causing the body morphology of zebrafish embryos to be curved. This effect is similar to that of Blebbistatin (HY-13813), but the animal mortality rate in the para-Nitroblebbistatin group is significantly reduced[2].
