Thiazole orange dimer YOYO 1 (YOYO 1) is a cell-impermeable cyanine dimer with no inherent fluorescence, and its fluorescence intensity increases significantly upon binding to double-stranded DNA (dsDNA). Thiazole orange dimer YOYO 1 can serve as a nuclear counterstain or a dead cell indicator (Ex/Em = 505/512 nm). In Vitro:Guidelines (The following is our recommended protocol, which serves only as a guideline and should be modified according to your specific needs.)
1. Prepare Working Solution
1.1 Dilute the stock solution with pre-warmed Hanks' solution containing 20 mM Hepes buffer (i.e., HH buffer), serum-free cell culture medium or PBS to prepare a working solution with a concentration of 1-10 μM.
Note: Adjust the concentration of the working solution according to actual conditions, and prepare it fresh before use.
2. Cell Staining
2.1 Culture the cells
2.2 Remove the cell culture medium and fix the cells.
2.3 Add 1-10 μM dye working solution, gently shake to completely cover the cells, and incubate for 15-60 minutes.
2.4 Aspirate the dye working solution, add HH buffer or other buffer to wash 2-3 times, 5 minutes each time.
2.5 Observe using a fluorescence microscope.
