Pleuromutilin (Drosophilin B) exhibits antibacterial activity through inhibition of protein synthesis[1].
In Vitro: Pleuromutilin exhibits board-spectrum antibacterial activity against Gram-positive bacteria (such as MRSA, S. pyogenes, S. pneumoniae), some fastidious Gram-negative bacteria (such as H. influenzae and M. catarrhalis), anaerobic bacteria (such as C. difficile, P. acnes), atypical pathogens (such as M. pneumoniae, C. pneumoniae), and mycobacteria (such as M. tuberculosis)[2].
Pleuromutilin binds to the peptidyl transferase center (PTC) of the 50S subunit of the bacterial ribosome, induces conformational changes in ribosomal RNA, interfers with the translocation of tRNA, blocks the translation process, thereby inhibiting the bacterial protein synthesis[2].
