NCDM-32B is a potent and selective KDM4 inhibitor, with IC50 values of 3.0 μM for KDM4A and 1.0 μM for KDM4C in in vitro enzyme assays. NCDM-32B specifically increases global H3K9me3/me2 levels in basal breast cancer cells. NCDM-32B impairs the viability of KDM4C-amplified basal breast cancer cell lines (HCC1954 and Colo824). NCDM-32B can be used for the study of breast cancer[1][2].
In Vitro:NCDM-32B (40 μM, 48 h) increases global H3K9me3/me2 levels without affecting H3K4me3/me2 levels or KDM4C protein level in HCC1954 breast cancer cells[1].
NCDM-32B (6.25-100 μM) impairs viability of HCC1954 and Colo824 basal breast cancer cells in a dose-dependent manner[1].
NCDM-32B shows potent KDM4 inhibition with IC50 values of 3.0 μM for KDM4A and 1.0 μM for KDM4C[1].
In Vivo:NCDM-32B (5.0 mg/kg, i.p., daily, from embryonic day 10.5 to 18.5) enables one BrafQ241R/+; Cre embryo to survive until postnatal day 21 (weaning age) in the mouse model[1].
