Introduction of 119725-20-1 :

Fupenzic acid is a triterpenoid compound that can be found in the fruits of plants in the genus Rubus. Fupenzic acid inhibits LPS (HY-D1056)-induced NF-κB activation and reduces the expression of NO, NOS-2, COX-2, inflammatory cytokines, and chemokines. Fupenzic acid binds to RBMS1, downregulates RBMS1/cGAS/STING signaling, enhances PINK1/Parkin-mediated mitophagy, and alleviates oxidative stress and radiation-induced liver fibrosis. Fupenzic acid exhibits cytotoxic activity against various tumor cells and can inhibit melanin production. Fupenzic acid has inactivation, protective, and replication-inhibitory activities against TMV and CMV. Fupenzic acid can be used in research related to liver fibrosis, malignant glioma, colon cancer, gastric cancer, hepatocellular carcinoma, TMV, and CMV infection[1][2][3][4][5]. In Vitro:Fupenzic acid (0-50 µM; 30 min) inhibits LPS (HY-D1056)-induced NO release in mouse peritoneal macrophages, with an IC50 of 16.7 µM[1].
Fupenzic acid (10-20 µM; 30 min) inhibits LPS-induced NOS-2 and COX-2 protein expression in mouse peritoneal macrophages[1].
Fupenzic acid (10-20 µM; 30 min) downregulates LPS-induced NOS-2, COX-2, IL-6, IL-10, IL-12, IL-18, KC, TNF-α, and IP-10 mRNA expression in mouse peritoneal cells[1].
Fupenzic acid (0-90 min) impairs IκBα/IκBβ degradation and reduces nuclear NF-κB p65 accumulation in LPS-activated mouse peritoneal macrophages[1].
Fupenzic acid (10-20 μM) downregulates RBMS1/cGAS/STING signaling in TGF-β1-stimulated LX-2 cells[2].
Fupenzic acid (20 μM) modulates the interaction of RBMS1 with cGAS and STING in LX-2 cells[2].
Fupenzic acid (10-20 μM) directly binds RBMS1 in LX-2 lysates and increases RBMS1 thermal stability in CETSA[2].
Fupenzic acid binds RBMS1 LBD in SPR with a KD of 0.51 μM[2].
Fupenzic acid (0.5-160 μM; 24 h) reduces LX-2 cell viability[2].
Fupenzic acid (10-20 μM; 24 h) inhibits TGF-β1-induced expression of fibrosis markers in LX-2 cells[2].
Fupenzic acid (10-20 μM; 24 h) attenuates TGF-β1-induced oxidative stress, inflammation, and mitochondrial dysfunction in LX-2 cells[2].
Fupenzic acid (20 μM; 24 h) inhibits cGAS-STING signaling and fibrosis/inflammation gene expression in LX-2 cells via RBMS1[2].
Fupenzic acid (48 h) shows selective cytotoxicity against HCT116, BGC823, and HepG2 cells, with IC50 values of 16.31 μM, 21.94 μM, and 23.40 μM, respectively[4].
Fupenzic acid does not inhibit α-glucosidase[4].
Fupenzic acid (100 mg/L) shows 25.11% TMV inactivation, 22.89% CMV inactivation, and 19.36% TMV protection in Nicotiana glutinosa/Chenopodium amaranticolor leaf assays[5].
Fupenzic acid (100 mg/L) inhibits TMV replication by 22.7640%[5]. In Vivo:Fupenzic acid (10-20 mg/kg/day; p.o. gavage; once daily for the first month, then once every 3 days from the second to the sixth month; total 6 months) ameliorates RT-induced liver fibrosis in C57BL/6 mice, whereas RBMS1 overexpression attenuates its ameliorative effects on liver injury, fibrosis markers, inflammation, oxidative stress, and mitochondrial dysfunction[2].
Fupenzic acid exhibits enhanced antitumor efficacy after gelation (FA-gel) in the U118-MG nude mouse model, accompanied by upregulation of caspase-3 and downregulation of Ki67 in tumor tissues[3].
