CDDO-dhTFEA (RTA dh404) is a synthetic oleanane triterpenoid compound which potently activates Nrf2 and inhibits the pro-inflammatory transcription factor NF-κB[1]. CDDO-dhTFEA restores hypertension (MAP), increases Nrf2 and expression of its target genes, attenuates activation of NF-κB and transforming growth factor-β pathways, and reduces glomerulosclerosis, interstitial fibrosis and inflammation in the chronic kidney disease (CKD) rats[2].
In Vitro:CDDO-dhTFEA (0-500 nM, 24 h) upregulates the expression of antioxidant enzymes such as HO-1, SOD, catalase, and GCLC, reduces H2O2-induced oxidative stress, and inhibits apoptosis in human pancreatic tissue[3].
CDDO-dhTFEA (0-500 nM, 24 h) reduces the production of cytokines and chemokines (IL-1β, Il-4, and IFN-γ) in human pancreatic tissue[3].
In Vivo:CDDO-dhTFEA (1 mg/kg, po, once daily for 3 days) protects pancreatic tissue, exhibits anti-inflamamtory activity in rats acute pancreatitis model[3].
