5 (6)-Carboxy-2',7'-dichlorofluorescein is a fluorescent probe substrate that acts on MRP1 and MRP3/Mrp3, passively diffuses across cell membranes, and is actively effluxed by MRP1 in MRP1-positive cells. 5 (6)-Carboxy-2',7'-dichlorofluorescein is transported across membranes in an ATP-dependent manner, but does not stimulate ATPase activity[1][2]. In Vitro:5(6)-Carboxy-2',7'-dichlorofluorescein (4 μg/mL; 4 h) is a specific substrate for MRP1 in A549, H358, and H322 cells, as its transport is inhibited by MK571[1]. In Vivo:5(6)-Carboxy-2',7'-dichlorofluorescein (1 μM; perfusion; single-pass; 35 min followed by 25 min CDF-free perfusion) exhibits measurable basolateral clearance in vehicle-treated isolated perfused rat liver and serves as a probe for Mrp3-mediated transport[2].
