NK-611 (VP 19) is an epipodophyllotoxin derivative. NK-611 induces DNA double-strand breaks by inhibiting topoisomerase II (IC50 = 56 μM). NK-611 does not inhibit microtubule polymerization, thus avoiding the side effects of the parent compound, Podofilox (HY-15552). NK-611 exhibits broad-spectrum antitumor activity and demonstrates potent efficacy in in vivo models of leukemia. NK-611 can be used in cancer research[1][2].
In Vitro:NK-611 (0.51-51 μM, 1 h- 28 d) exhibits potent cytotoxicity against L1210 cells with an IC50 of 0.33 μM and also exhibits time-dependent broad-spectrum anti-tumor activity against 17 different types of tumors, including renal cell carcinoma and colorectal cancer[1][2].
In Vivo:NK-611 (i.p., single dose) exhibits significant anti-tumor activity in a mouse leukemia model with an ED50 of 0.9 mg/kg[1].
